  Characterization and Identification Of Mercury-resistant Bacteria From Kalimas River Surabaya-Indonesia By Numerical Phenetic Taxonomy (Indexed in THOMSON)

Abstrak : Kalimas River in Surabaya, East-Java Indonesia is directly connected to the Tanjung Perak Harbor. The river was known to be contaminated by mercury between 0.105 to 6.38 mg/L. Mercury-resistant bacteria are able to survive under extreme condition of mercury contamination. This present work was aimed to isolate and to characterize mercury-resistant bacteria from water and sediments of the Kalimas River using a 25 mg/L HgCl2- containing nutrient agar. The major task of study was to develop a mercury bioremediation agent. The bacterial isolates were characterized by utilizing a numerical phenetic taxonomy method based on 100 phenotypic characters, including morphological, biochemical and physiological characters. The results showed that the bacterial isolates were not only resistant to 25 mg/L HgCl2, but also could survive on 5% NaCl, as well as on a 25 mg/L of PbCl2, CdCl2, and CuCl2-containing nutrient agar. All the bacterial isolates were catalase positive with three distinguished morphology of cocci, ovoid, and rods. Based on the numerical phenetic taxonomy analysis the isolates separate into three different phenotypic groups at similarity level of 72-77%. The first group containing isolates S1, S13, SS19, DA11 were identified Bacillus along with two reference strains of Bacillus cereus ATCC1178 and B. subtilis ATCC6633. Second group containing A3 was identified Staphylococcus together with the reference strain of Staphylococcus aureus ATCC6538P. The single membered group (S8) was identified to be member of the genus Azotobacter.
